氣相色譜(GC)與液相色譜(LC)的比較

氣相色譜和液相色譜相比各有什么特點呢?讓我們從以下幾個方面進行考察:
一、活動(dong)相
GC用(yong)氣體作(zuo)活動相(xiang),又叫載(zai)氣。常用(yong)的(de)(de)載(zai)氣有氦氣、氮氣和氫(qing)氣。與LC相(xiang)比,GC活動相(xiang)的(de)(de)種類(lei)少(shao),可(ke)選擇范(fan)圍小,載(zai)氣的(de)(de)主要作(zuo)用(yong)是將樣品帶進GC系統進行(xing)分離(li),其本(ben)身對(dui)分離(li)結(jie)果的(de)(de)影響(xiang)很(hen)有限。而(er)在(zai)LC中,活動相(xiang)種類(lei)多,且對(dui)分離(li)結(jie)果的(de)(de)貢獻(xian)很(hen)大。換一(yi)(yi)個角度看,GC的(de)(de)操縱參數優化相(xiang)對(dui)LC要簡單(dan)一(yi)(yi)些。此外,GC載(zai)氣的(de)(de)本(ben)錢要低(di)于(yu)LC活動相(xiang)的(de)(de)本(ben)錢。
二、固定(ding)相
由(you)于GC的載(zai)氣(qi)種(zhong)(zhong)(zhong)類相(xiang)(xiang)(xiang)對少,故其(qi)分(fen)離(li)(li)(li)(li)選擇(ze)性(xing)(xing)主要(yao)(yao)通過不同(tong)的固(gu)定(ding)(ding)相(xiang)(xiang)(xiang)來改變,尤其(qi)在(zai)填充柱GC中(zhong),固(gu)定(ding)(ding)相(xiang)(xiang)(xiang)常由(you)載(zai)體和(he)(he)涂敷在(zai)其(qi)表(biao)面(mian)的固(gu)定(ding)(ding)液組(zu)成(cheng),這對分(fen)離(li)(li)(li)(li)有(you)決定(ding)(ding)性(xing)(xing)的影響,所以,導致了種(zhong)(zhong)(zhong)類繁多的GC固(gu)定(ding)(ding)相(xiang)(xiang)(xiang)的開發研(yan)究。迄今(jin)已有(you)數(shu)百種(zhong)(zhong)(zhong)GC固(gu)定(ding)(ding)相(xiang)(xiang)(xiang)可供(gong)我(wo)們選擇(ze)使用,但常用的LC固(gu)定(ding)(ding)相(xiang)(xiang)(xiang)也就十幾種(zhong)(zhong)(zhong)。故LC在(zai)很大(da)程度(du)上(shang)要(yao)(yao)靠(kao)選用不同(tong)的活(huo)動相(xiang)(xiang)(xiang)來改變分(fen)離(li)(li)(li)(li)選擇(ze)性(xing)(xing)。當然,毛細管(guan)GC常用的固(gu)定(ding)(ding)相(xiang)(xiang)(xiang)也不過十幾種(zhong)(zhong)(zhong)。在(zai)實際分(fen)析(xi)中(zhong),GC一般是(shi)選用一種(zhong)(zhong)(zhong)載(zai)氣(qi),然后通過改變色(se)譜(pu)柱(即固(gu)定(ding)(ding)相(xiang)(xiang)(xiang))以及操縱參(can)數(shu)(柱溫順載(zai)氣(qi)流速等(deng))來優化分(fen)離(li)(li)(li)(li),而LC則往(wang)(wang)往(wang)(wang)是(shi)選定(ding)(ding)色(se)譜(pu)柱后,通過改變活(huo)動相(xiang)(xiang)(xiang)的種(zhong)(zhong)(zhong)類和(he)(he)組(zu)成(cheng)以及操縱參(can)數(shu)(柱溫順活(huo)動相(xiang)(xiang)(xiang)流速等(deng))來優化分(fen)離(li)(li)(li)(li)。
三、分析對(dui)象
GC所能直接分(fen)(fen)離(li)的樣(yang)品是可(ke)揮發、且熱穩定的,沸(fei)點一(yi)般不(bu)超過500℃。占(zhan)有關資料統計,在目前已知(zhi)的化合(he)物中,有20%~25%可(ke)用(yong)(yong)GC直接分(fen)(fen)析,其(qi)余原則上均可(ke)用(yong)(yong)LC分(fen)(fen)析。也就(jiu)是說GC的分(fen)(fen)析對象遠沒有LC多
